Adjuvant potential of archaeal synthetic glycolipid mimetics critically depends on the glyco head group structure.

Subunit vaccines capable of providing protective immunity against the intracellular pathogens and cancers that kill millions of people annually require an adjuvant capable of directing a sufficiently potent cytotoxic T lymphocyte response to purified antigens, without toxicity issues. Archaeosome lipid vesicles, prepared from isoprenoid lipids extracted from archaea, are one such adjuvant in de...

Metabolism and Function of Bacterial Lipids

Relatively little is known about the intermediary metabolism of bacterial phospholipids. Indeed, as Assilineau and Lederer have pointed out in their authoritative review (l), comparatively few bacterial phospholipids have been well characterized chemically by modern techniques. Interest in the chemistry and metabolism of bacterial lipids is, however, rapidly growing. In the past few years, publ...

Non-bilayer lipids stimulate the activity of the reconstituted bacterial protein translocase.

To determine the phospholipid requirement of the preprotein translocase in vitro, the Escherichia coli SecYEG complex was purified in a delipidated form using the detergent dodecyl maltoside. SecYEG was reconstituted into liposomes composed of defined synthetic phospholipids, and proteoliposomes were analyzed for their preprotein translocation and SecA translocation ATPase activity. The activit...

Adjuvant activity of Mycobacterium leprae.

Mycobacterium leprae organisms isolated from infected spleen and liver tissue by zonal centrifugation were shown to possess adjuvant activity. Histochemical examination of the footpad macrophage epithelioid granuloma showed that macrophages contained large amounts of hemosiderin after the injection of M. leprae and much smaller amounts after the injection of M. tuberculosis.
